| Аннотация: | OBJECTIVE: By constructing cochlear hair cell recombinant Prestin adeno-associated virus-2 (AAV-2) vector, it provides a tool for the study of cochlear Prestin protein regulation. METHODS: The guinea pig Prestin gene was used as a template for PCR amplification and extraction. The AAV-2-Prestin vector was constructed by genetic engineering. The expression level of Prestin was detected by Western blot on the 1st, 3rd and 5th day after transfection of cultured cochlear hair cells. RESULTS: After transfecting HEK-293T cells with pAAV-EGFP-Prestin proviral plasmid, the expression of weak green fluorescent protein was observed on the 1st day, and the expression of green fluorescent protein was peaked on the 3rd day. Conclusion: The recombinant adeno-associated virus-2-Prestin vector was successfully constructed and has good transfection efficiency and persistence. |
